Sleep for resilience: Translational insights from mouse models to human dementia with Brianne Kent
Sleep disturbance is a common symptom of Alzheimer’s disease (AD) and reduced slow-wave sleep (SWS) is linked to increased risk of dementia. Our translational work spanning mouse models and human cohorts is evaluating sleep as a modifiable target for promoting resilience to dementia. In a mouse model of AD, chronic administration of the SWS-enhancing antidepressant trazodone reduced markers of AD neuropathology, in addition to consolidating sleep and improving short-term and long-term memory. In humans, we have been studying how sleep is associated with cognitive performance and dementia risk in the Brain Resilience Study, a new open-science initiative integrating sleep, cognition, neuroimaging, and psychosocial data across 1,000+ older adults to advance early, personalized, and equitable dementia prevention.
